چکیده

Ring canals connecting Drosophila germline, follicle and imaginal disc cells provide direct contact of cytoplasm between cells. To date, little is known about the formation, structure, or function of the somatic ring canals present in follicle and imaginal disc cells. Here, we show by confocal and electron microscopy that Pavarotti kinesin-like protein and Visgun are stable components of somatic ring canals. Using live-cell confocal microscopy, we show that somatic ring canals form from the stabilization of mitotic cleavage furrows. In contrast to germline cells, syncytial follicle cells do not divide synchronously, are not maximally branched and their ring canals do not increase in size during egg chamber development. We show for the first time that somatic ring canals permit exchange of cytoplasmic proteins between follicle cells. These results provide insight into the composition and function of ring canals in somatic cells, implying a broader functional significance for syncytial organization of cells outside the germline.

Long-term live imaging provides new insight into stem cell regulation and germline-soma coordination in the Drosophila ovary.

The Drosophila ovariole tip produces new ovarian follicles on a 12-hour cycle by controlling niche-based germline and follicle stem cell divisions and nurturing their developing daughters. Static images provide a thumbnail view of folliculogenesis but imperfectly capture the dynamic cellular interactions that underlie follicle production. We describe a live-imaging culture system that supports ...

Localization and functions of Protein Kinase A during Drosophila oogenesis

We have characterized the requirements for the Protein Kinase A (PKA) catalytic subunit, DC0, in Drosophila oogenesis. Intercellular bridges in egg chambers from PKA deficient females are unstable, leading to the formation of multinucleate nurse cells by fusions of adjacent cells. Germline clones of cells homozygous for null mutations of DC0 indicate that PKA acts autonomously in the germline. ...

Genetic analysis of the actin cytoskeleton in the Drosophila ovary.

The Drosophila ovary provides a favorable model system in which to study cellular morphogenesis. The development of a mature egg involves a syncytium of 16 germline cells and over 1000 somatically derived follicle cells. Intercellular transport, stable intercellular bridges, cell migrations, cell shape changes, and specific subcellular localization of many embryonic patterning determinants cont...

Antisera against a channel-forming 16 kDa protein inhibit dye-coupling and bind to cell membranes in Drosophila ovarian follicles.

In Drosophila ovarian follicles, communication via gap junctions can be observed between the oocyte and its surrounding follicular epithelium. In the present study, the intercellular exchange of the fluorescent tracer Lucifer Yellow was analysed following pressure-injections of five different sera or protein solutions into the oocyte of stage-10 follicles. Three of the tested sera are directed ...

Intercellular communication between germ line and somatic line is utilized to control the transcription of ZAM, an endogenous retrovirus from Drosophila melanogaster.

ZAM is an long terminal repeat (LTR) retrotransposon from Drosophila melanogaster that bears striking resemblance to the vertebrate retroviruses, in their structure and replication cycle. This element transposes via an RNA intermediate and its reverse transcription, and ultimately inserts copies within the germ line. In this paper, we show that intercellular communication established between th...
